Near its eastern terminus, NY 52 originally followed present day NY 216 and NY 292 between Stormville and Towners. The alignment moved to its current location in 1937.

Before the Newburgh-Beacon Bridge opened to traffic in 1963, a ferry serviced NY 52 across the Hudson River. NY 52 followed Plank Rd. and South St. to the western terminal in Newburgh. From the eastern terminal at Riverfront Park in Beacon, NY 52 picked up Main St. (Current Business NY 52) and followed that east before rejoining the current alignment near Fishkill. NY 52 moved to its current alignment upon completion of the Newburgh-Beacon Bridge.
