NY 85's original western end was originally at NY 81 in Oak Hill and followed Makley and Seifert Roads to Medusa and Albany CRs 351, 402, 404, and 401 to Westerlo, and NY 143 before picking up the current alignment. It was realigned in 1936 along Albany CRs 352, 360, 359, and 353, from Medusa to Rensselaerville and truncated to its current location in Rensselaerville two years later (The western extension shown on some maps as late as 1971).

At the east end, NY 85 was originally assigned along New Scotland Rd. and New Scotland Ave. into Albany before ending at US 20 at Washington Park. NY 85 was gradually shifted onto its current expressway and super-2 alignment from the 1960s through 2007.
